Research Interests
Inflammatory signals, including exogenous mediators such as lipopolysaccharide and endogenous mediators such as cytokines, stimulate cellular phospholipase (PLA2) activities resulting in the release of arachidonic acid from membrane phospholipid stores. Arachidonic acid is converted into prostaglandins, prostacyclins and thromboxanes by the cyclooxygenase (COX) pathway or leukotrienes and hydroxyeicosatetraenoic acids (HETEs) by the lipoxygenase (LO) pathway. Prostaglandins and other derivatives of arachidonic acid metabolism, are involved in a number of physiological and pathophysiological phenomenon including: acute and chronic inflammation, cardiovascular disease, immune response, arthritis, inflammatory bowel disease, respiratory disorders and colon cancer. My laboratory is interested in understanding the expression and regulation of enzymes involved in arachidonic acid metabolism, especially, cyclooxygenases, lipoxygenases and phospholipases and their role in the development of cardiovascular diseases such as atherosclerosis. Currently, my laboratory is looking at the role of cyclooxygenases in atherosclerotic lesion formation using animal models. Atherosclerosis Research Unit (ARU) at UCLA identified a series of biologically active oxidized lipids, derived from an arachidonic acid containing phospholipid; palmitoyl arachidonyl phosphatidyl choline (PAPC). These oxidized lipids are potent mediators of monocyte adhesion and migration, which is an early event in the development of atherosclerotic lesions. In collaboration with ARU faculty, we completed a screen to identify genes, whose expression is regulated by the biologically active oxidized lipids in human aortic endothelial cells. We have cloned a number of interesting genes and are currently in the process of characterizing some of thege genes and determining their role in the development of atherosclerotic lesions in animal models.
For more detailed information, please visit the lab website.
